Temperature sensitive library (ts1-ts1056)
Temperature sensitive library was constructed. Cells that were mutagenised with EMS were screened. The colonies which grew at 30 degree, but not at 40 degree on YE agar plates were isolated. Here we present the information of damage sensitivity of each strains.
(1) Damage Sensitivity (view the table)
Either overnight culture (x1) or diluted*1 culture (x1/10) were spotted*2 onto YE plates that contain DNA damage agents. 12 mM, 25 mM or 50 mM were used for HU (Hydroxyurea). 0.05 uM, 0.2 uM or 0.5 uM were used for CPT.
        3: Well grown colonies
        2: Grown colonies with red colours on phloxin B containing plates.
        1: tiny colonies with Red colours.
        0: No formation of colonies.
*1 Approximately.
*2 Approximately 5 uL of culture were spotted by hedgehog.
(2) DAPI Phenotypes(view the photo )
Cells were cultivated to early log phase in YEAU and shifted up to 40Ž for 3 hrs.Then cells were fixed by 2.5% glutaraldehyde solution for 30 min, washed with PBS(pH7.4) and stained by 25 ƒĘg/ml DAPI solution.
