Affected Gene uap1
Origin and Depositor University of Yamanashi (Atsuo Kawahara)
Link to ZFIN
Information
There is a deletion of 9 bp and an insertion of 2 bp (7 bp deletion in total) within uap1 gene as a result of genome editing using the CRISPR/Cas9 system. The deletion results in frame shift and nonsense mutation of the gene and generates a truncated protein with 73 amino acids (wild-type uap1 protein has 504 amino acids).
Insertion/deletion region of uap1 gene (small letters indicate insertions, hyphens indicate deletions):
wild-type: 5'-CTTCTTTAAGTCGGCCATGAAGACGTCTGGGCAGAGCT-3'
mutant: 5'-CTTCTTTAAGTC-------tgGACGTCTGGGCAGAGCT-3'
The following primers can be used to detect this mutation:
Forward: 5'-AGATGAGTCTCGACCTGGA-3'
Reverse: 5'-GTGTCCACCTTCTCCTGTGA-3'
This mutation causes the following recessive phenotypes: shorter protrusion of the mouth at 7 dpf and larval death before 14 dpf (no homozygous mutant animal has been found after 14 dpf).
This mutant may have a transgene for lyz:EGFP.
