The kdm2banf7 zebrafish mutant was developed using CRISPR/Cas9 technology. The kdm2banf7 mutant carrys a 9-bp deletion and a 2-bp insertion in the exon 4 of kdm2ba, creating a premature stop codon as described below. Wild type and mutant alleles can be determined by BspEI digestion of a PCR product generated with the following primer set: 5'-CCCAACAAACCCAAACCTAA-3'and 5'-AGAAGAATCACTCCCTGCCC-3'(Wt, 106-bp and 460-bp; Mutant, 566-bp). The Tg(fli1a:LIFEACT-mCherry)ncv7Tg line has been developed and deposited by Dr. Naoki Mochizuki (National Cerebral and Cardiovascular Center Research Institute). |
