| This line has heterozyogous mutant alleles that lacked 10 bp at exon2 and 16 bp at exon3 in lats1 and lats2 gene, respectively. To genotype this mutation, we used primer pairs for amplification: lats1 (5’-GGCACTTAACATATGCTTTTACATG-3’and 5'- TTTGCTGCTGTCTGCGGAGCTGTT-3') and lats2 (5'-AGAGTTTGTGTGAGAGAAAACAGG-3' and 5'-GCATTGACCAGATCCTGTAGCATC-3'). This line expresses a Nuclear localized signal (Nls)-tagged Eos Fluorescent protein (EosFP) under the control of myh6 atrial cardiomyocyte promoter and Nls-mCherry under the control of myl7 promoter. |
