DsRed fluorescence is driven by pax7a promoter. Once recombination is induced by cre at the loxp sites, sox5 (WT) becomes expressed there. For the purpose of establishing this strain, TgBAC(pax7a:loxp-dsred-loxp-sox5^WT-2A-venus) DNA was injected into embryos of ml-3 mutant with B/B (having normally-melanized melanophores), which had been obtined by crossing ml-3 original b/b mutant (not having normally-melanized melanophores, since b allele is a recessive loss-of-function mutant allele) with Nagoya (B/B). Thus this strain has copies of TgBAC transgene as well as dominant B alleles. |
Notes | Please note that we will use OK-Cab (MT830) for artificially insemination with frozen sperm of this strain. |

TG1210_sox5^ml-3ml-3 TgBAC(pax7aloxp-dsred-loxp-sox5^WT-2A-venus) In the absence of cre mRNA, sox5-/-;Tg embryos (sox5-/-;TgBAC(pax7a:loxp-dsred-loxp-sox5^WT)) were strongly expressing Dsred (upper panels, no injection). When cre mRNA was injected in the 1-cell sox5-/-;Tg embryos, they became positive for Venus (lower panels, injection with Cre mRNA). Venus-expressing animals showed rescued xanthophore and leucophore phenotypes in sox5-/- mutants. |
