Allele jt7
Strain Name atg2a, atg2b
Type Mutant
Genotype
Affected Gene atg2a, atg2b
Origin and Depositor The University of Tokyo (Noboru Mizushima)
Link to ZFIN
Information
atg2a- and atg2b-double mutants (loss-of-function mutation of both atg2a and atg2b) were generated by CRISPR/Cas9.

50-base deletion was introduced in exon3 of atg2a gene by using CRISPR method.
The heterozygous fish was identified by heteroduplex mobility assay using followed primers:
atg2a-f, TGTGCAAACATTTCCAGCCCCTCCAG
atg2a-r, CTCACTGGTTCTGTATTTGGGTCGGC
Mutation:
5’-CATACAGGATGACCACCCTTGGG-3’ Wild-type allele
5’------------------CTTGGG-3’ Mutated allele (-50 bp)

5-base deletion was introduced in exon6 of atg2b gene by using CRISPR method.
The heterozygous fish was identified by heteroduplex mobility assay using followed primers:
atg2b-f, GGTTCCAACTTGGAGAAAGCTTGGG
atg2b-r, CAACATCCATCAGCCCACTACATTTGCGC
Mutation:
5’-AAACTCATCCCAGAACAGAGAGG-3’ Wild-type allele
5’-AAACTCATCCCAGA-----GAGG-3’ Mutated allele (-5 bp)
