This line expresses a FRET-based caspase sensor, YFP-DEVD-CFP, specifically in endothelial cells by fli1a promoter. In YFP-DEVD-CFP, CFP is fused with YFP by a caspase-sensitive amino acid peptide (DEVD). Upon induction of apoptosis, FRET is interrupted due to cleavage of the linker by caspase-3. Therefore, loss of FRET becomes a direct indicator of caspase activity. |
