Phenotypes of the kyo mutant. (A,B) Morphology and (C,D) Wright-Giemsa staining of the blood cells in the 4dpf wild-type (A,C) and kyo (B,D) mutant embryos. (E-H) o-Dianisidine staining for hemoglobin in the 4dpf wild-type (E) and kyo (F) mutant embryos. (G,H) At higher magnification, the enlarged blood cells were clearly seen in the blood vessels of the kyo mutant (H). The kyo mutant contained huge blood cells with a wrinkled surface (A-D). The major axis length of the blood cells was 24±5μm in the kyo mutant embryos, in contrast to 7±1μm in the wild-type embryos. Staining with o-dianisidine showed that the peroxidase activity was relatively normal, suggesting that hemoglobin synthesis was not impaired in the mutant (E-H). Blood cells isolated from the 4dpf wild-type (I,J) and kyo mutant (K,L) embryos were stained with propidium iodide. Fluorescence (I,K) and light (J,L) microscope images are shown. As a result, we found that the mutant blood cells had a single large nucleus (K,L).
