Characteristics The col10a1:nlGFP transgenic embryos express GFP in osteoblast progenitors and pre-mature osteoblasts (see Renn et al., Dev Biol 2013). At later stages, nlGFP expression is also found in chondrocytes. Note that the nuclear localization signal (nl) is leaky and GFP expression is also observed in cytoplasm.

A 5.865kb upstream regulatory region of the medaka col10a1 locus was amplified from genomic medaka DNA using the primers 5′.CCCATGCTTGTTTTCCTGTT-3′ and 5′.TCCAAACAGATCT- GACCTTGC-3′, and cloned in to a meganuclease vector with GFP followed by a nuclear localization signal (for details see Renn et al., Dev Biol 2013).

nlGFP expression in a Medaka embryo at 15 dpf. nlGFP expression can be found in cranial skeletal elements, around the centra of the vertebral bodies, in the neural and hemal arches, and in the fin rays of the caudal fin. For more details, please see Renn et al., Dev Biol 2013.
