Characteristics npba-expressing neurons are labeled with GFP (hrGFPII). Neuronal cell bodies labeled with GFP can be found in some brain regions, including the ventral preoptic area, habenula, and hypothalamus, in both sexes and in the dorsal preoptic area and ventral telencephalon only in females. GFP-labeled axons can be found in some regions, including the hypothalamus and pituitary, in both sexes and in the ventral medulla oblongata and spinal cord only in females. The specificity of GFP expression has been confirmed by double in situ hybridization detecting GFP and endogenous npba transcripts, which demonstrated that the distribution of the two transcripts almost completely overlapped (Hiraki-Kajiyama et al., 2019, eLife, 8:e39495). Results from only d-rR-Tg(npba-GFP)2 have been published in the papers (Hiraki-kajiyama et al., 2019, eLife, 8:e39495; Kikuchi et al., 2019, Endocrinology 160:827-839), but the same results have been obtained from d-rR-Tg(npba-GFP)1. No phenotypic difference has been seen between the d-rR-Tg(npba-GFP)1 and d-rR-Tg(npba-GFP)2 strains.

Structure of the transgene in d-rR-Tg(npba-GFP).
A 22-bp sequence containing the translation initiation site of npba in a medaka BAC clone (clone ID: ola1-180I09) was replaced by a 2,136 bp DNA cassette containing the humanized Renilla reniformis GFP II-coding sequence (hrGFPII; Stratagene, San Diego, CA), bovine growth hormone polyadenylation signal (BGH pA), and kanamycin resistance gene (Km).

Some brain regions, including the ventral preoptic area (vPOA) and dorsal medulla oblongata (dMO), and the pituitary (Pit) of both sexes show GFP fluorescence, while other regions, including the dorsal preoptic area (dPOA), ventral medulla oblongata (vMO), and spinal cord, show fluorescence only in females.
