A 10 bp insertion was generated by TALEN method in exon 1 of sox10a gene on LG1. The target sequences of TALEN were specific target: 5'-TTCTGAGTCGGAGCTGAGC-3' (left, sense) and 5'-TGAGAGTGAGTGACCGTCC-3' (right, antisense). The sox10a gene encodes a HMG type transcription factor SoxE family, and is expressed in the neural crest cells. Sox10a plays an important role for pigment cell develpment. The homozygous larvae show reduction in melanophore, iridophore and xanthophore numbers, and do not survive beyond hatching. The original background is Nagoya, with wild-type body color. |
