(G,H) At higher magnification, the enlarged blood cells were clearly seen in the blood vessels of the kyo mutant (H). The kyo mutant contained huge blood cells with a wrinkled surface (A-D). The major axis length of the blood cells was 24±5μm in the kyo mutant embryos, in contrast to 7±1μm in the wild-type embryos. Staining with o-dianisidine showed that the peroxidase activity was relatively normal, suggesting that hemoglobin synthesis was not impaired in the mutant (E-H). Blood cells isolated from the 4dpf wild-type (I,J) and kyo mutant (K,L) embryos were stained with propidium iodide. Fluorescence (I,K) and light (J,L) microscope images are shown. As a result, we found that the mutant blood cells had a single large nucleus (K,L).
