Comment PGCs formed tight bilateral clumps in wild-type embryos at st. 27. In yanagi (yanj6-29C) mutant embryos, however, PGCs did not coalesce but were loosely aligned in bilateral rows in the region more anterior than normal PGC clusters. The positioning of lateral line organs were also affected in this mutant embryos (Yasuoka et al.,MOD121:729-738). Homozygotes of yan were viable and fertile.

Whole-mount in situ hybridization using the olvas probe at st. 27. In the wild-type embryo (top), PGCs are clustered at the ventrolateral regions of the trunk (indicated by black arrowheads) at the axial level where the posterior trunk detach from the yolk. In the mutant embryo (bottom), PGCs are dispersed along the bilateral lines (broken lines) anterior to the sites of normal PGC clustering. White arrowheads indicate the pigment cells.
