TG1361_Tg(fli1DsRed)_transgene map For creating the fli1 promoter DsRed transgenic medaka, Tg(fli1:DsRed), the 4.6-kb upstream region of the medaka fli1 gene was amplified by using the following primers: fli1 promoter F-XhoI, 5-TGTGTCCTGAATGTCCTGAC-3; and fli1 promoter R-KpnI,5-TTTGCGCAAATTTTCGCGGACC-3. The digested fragment with XhoI and KpnI was subcloned into the XhoI/KpnI site of the I-SceI backbone vector, in which MCS/DsRed Express/SV40polyA of pDsRed Express (Clontech) was inserted into HindIII and AflII sites of PBSKI2 (AMAGEN). |
