Characteristics Fish of this strain grow fast and big, because of ectopic overexpression of the growth hormone. The transgene also expresses GFP as a reporter. Transgene: Medaka beta-actin promoter was ligated to Medaka growth hormone-IRES-GFP fusion gene in pGEM-T easy vector (Promega). An internal ribosome entry site (IRES)” from the pIRES-hrGFP-1a vector (Stratagene), “a GFP open-reading frame” from the phrGFP II-1 vector (Stratagene), and “SV40 polyA signals” from the pDsRed2-1 vector (Clontech).
Notes Females are severely sterile. Males are incompletely sterile. Therefore, this line should be maintained by corssing heterozygous males with ci females. We use OK-Cab (MT830) embryos in artificially- insemination. Please note that we will use OK-Cab (MT830) for artificially insemination with frozen sperm of this strain.
